Wet age-related macular degeneration (wet AMD) is a progressive retinal disease characterized by abnormal neovascularization in the macula, leading to significant vision loss or blindness. While intravitreal anti-VEGF therapies such as aflibercept are the current standard of care, 15–40% of patients exhibit incomplete or no response, and frequent injections are associated with risks such as infection, retinal detachment, and patient non-compliance.

 

SCAI-010 is a novel, non-invasive topical formulation of cabozantinib, developed using our proprietary MOASIS®

technology to enable targeted delivery to the retina. Cabozantinib is a multi-target TKI with high potency against VEGFR2, as well as additional activity against MET and AXL, which are implicated in angiogenic resistance, inflammation, and fibrosis.

Compared to VEGFR-selective TKIs like axitinib, cabozantinib offers a broader mechanism of action that may improve efficacy in refractory or inflammation-driven AMD.

 

Importantly, cabozantinib’s long systemic half-life (~99 hours) suggests potential for extended dosing intervals, a key advantage in reducing treatment burden.
